M3 inhibition of CCL19- and CCL21-induced [³⁵S]GTPγS exchange in Ba/F3-hCCR7 membranes. Ba/F3-hCCR7 membranes were incubated for 60 min at room temperature in GTPγS binding buffer (as described in Materials and Methods) containing 10 μM GDP in the presence or absence of 10 nM CCL19 or CCL21 and the indicated concentrations of rM3. Following the addition of 0.3 nM [³⁵S]GTPγS, the incubation continued for a further 60 min. Membrane-associated radioactivity was measured using SPA technology. Data represent the mean total binding and standard errors of the means (error bars) of triplicate determinations from a representative experiment (n = 3).
