Histone methyltransferase activity was assayed by incubation of free histones in the presence of S-adenosyl-l-[methyl-³H]methionine, and incorporated radioactivity was determined by filter binding. The GST pull-down experiments were done as previously described (17, 19) with GST fusions to the N-terminal 57 residues of human H3 (wild type [wt]) and point mutants, and 20% of the total reaction mixture was loaded as input.
