Under-replication in dnaAT174P, dnaAA345S and dnaNG157C mutants. (A) Mutations dnaAT174P, dnaAA345S and dnaNG157C lead to reduced chromosome content. Cells (DHB4, SMG275, SMG217 and SMG219) grown exponentially were stained and analyzed by flow cytometry. Average cell mass and DNA contents were derived from the flow cytometry histograms (see Materials and methods). The average numbers from three experiments are given. (B) Cells growing exponentially in AB glucose-CAA medium with uridine (100 μg/ml) were treated with rifampin and cephalexin to block initiation but allow elongation. Distinct peaks represent the accumulation of cells with integral numbers of chromosomes that reflect the numbers of origins at the time of drug action.
