Undeleted strains bound to an undeleted probe migrate low in the gel, while deleted strains migrated high in the gel because of the large malformation of the DNA helix. In contrast, deleted strains bound to a deleted probe migrated more according to size due to relatively little mismatch, while undeleted strains migrated high in the gel. PCR-amplified control strains B958, Ch2, AL, NC, Med−, and Med+ were denatured with and reannealed to (A) the undeleted Ch2 probe and (B) the deleted Med+ probe.
