Inhibition of calcineurin by cyclosporin A had no effect on resting cycling of NFATc1-GFP in cultured adult mouse FDB muscle fibers. (A) CsA (1 μM) treatment for 2 h had no effect on the subcellular localization of NFATc1-GFP (6 nuclei from 6 fibers). (B) A fiber expressing NFATc1-GFP is shown at the start treatment of 1 μM CsA (0 min), after 30 min of CsA (the start of 40 nM LMB), and after 90 min of CsA (and 60 min of LMB) treatment, showing that in the presence of CsA, LMB still caused nuclear accumulation as well as nuclear body formation of NFATc1-GFP.
