Effect of treatment duration and inhibitor concentration on replicon RNA level. Replicon-bearing Huh-7 cells (clone 16) were incubated for 14 days in media (without G418 selection) containing 0.5 μM (1.2 × 90% inhibitory concentration), 2.5 μM (6 × EC₉₀) SCH 503034, or 5 μM (12 × EC₉₀) SCH 503034. Compound and medium were refreshed daily. HCV replicon RNA level was determined by TaqMan as described in Materials and Methods and normalized to time zero. Experiments were performed in triplicate, and error bars represent the standard deviations.
