In vitro dephosphorylation of Wzc by Wzb. Purified His-tagged Wzc was incubated at 30°C with purified Wzb as described in Materials and Methods. The reaction was stopped at 0, 30, and 60 min and 12 h, and aliquots were analyzed by Western immunoblotting using antiphosphotyrosine antibodies (PT66). As a control Wzc was incubated without Wzb. The following lanes were incubated with Wzb for the indicated times: lane 1, 0 min; lane 2, 30 min; lane 3, 60 min; lane 4, 12 h.
