MinD ATPase assays. (A) Stimulation of H-MinD ATPase activity by phospholipid and MinE-H. Proteins (each at 12 μM) were added to RB containing 1 mM [α-32P]ATP (0.33 μCi/nmol) and 0.5 mg of phospholipid vesicles/ml as indicated. Mixtures were placed at 30°C, and reactions were started by the addition of MgCl2 to 5 mM. The amounts of ADP produced after 40 min were used to calculate specific activities. D, H-MinD; E, MinE-H; *E, *MinE-H; L, phospholipid vesicles. (B) Nucleotide competition. H-MinD (5 μM) was added to RB containing 100 μM [α-32P]ATP (3.3 μCi/nmol) and a 1 mM concentration of the indicated unlabeled nucleotide. Mixtures were placed at 30°C, MgCl2 was added to 5 mM, and the amounts of ADP produced after 60 min were determined.
