COL was grown in TSB, and the COLspacmurF conditional mutant was grown in TSB supplemented with the following IPTG concentrations: 100 μM, 10 μM, 7.5 μM, 5 μM, and 2.5 μM. Samples were taken at an OD620 value of 0.7 and assayed for murF transcription by Northern blotting.
