Photographs of electrophoretically separated cell lysates reacted with antibody against total PKB/Akt, phosphorylated PKB/Akt serine 473 or threonine 308, U_S3, or β-actin. HEp-2 cells were mock infected or exposed to 10 PFU of HSV-1(F), ΔU_S3, or ΔICP4 per cell and maintained in DMEM supplemented with 5% NBCS. The cells were harvested at the time points shown, solubilized, and normalized for protein content. Equal amounts of protein lysates (120 μg/lane) were subjected to electrophoresis on a denaturing gel, transferred to a nitrocellulose sheet, and reacted with antibodies as indicated.
