Amounts of Src remain unaltered in cells infected with wild-type or mutant HSV viruses. Replicate HEK293 or HEL cells in T25 flasks were mock infected or exposed to 10 PFU of HSV-1(F), ΔICP0, or ΔUL41 (lacking the gene encoding the virion host shutoff protein [vhs]) viruses per cell. The cells were harvested 4, 8, and 12 h after infection. The cells were lysed as described in Materials and Methods, and 50 μg of total cell lysate from each sample was electrophoretically separated in a denaturing polyacrylamide gel, transferred to a nitrocellulose sheet, and reacted with anti-Src antibody.
