HSV infection enhances the phosphorylation of Tyr416 of Src. To detect tyrosine phosphorylation specific to just Src kinase, equal amounts of whole-cell lysate were reacted with anti-Src for 3 h at 4°C. One-third of the immune precipitates were solubilized, electrophoretically separated in a denaturing gel, and reacted with anti-Src-pY416 or anti-Src-pY527. The blots were also stripped and reprobed with anti-Src to determine the total amount of Src protein that was immune precipitated. The reactive protein bands were quantified with the aid of a General Dynamics Storm 860 phosphorimager or exposed to X-ray film.
