Northern blotting of total RNA isolated from Δ25 cells carrying plasmids pRS426 or pRS426-U6 (+U6). Cells were grown at 25°C, then shifted to 37°C for 2 h. Total RNA was isolated and fractionated on a 5% nondenaturing polyacrylamide gel for Northern blotting and probed sequentially with U4, then U6.
