The KIR receptor has a lysine close to the center of the TM domain (position 9), while the FcαRI receptor carries an arginine in the N-terminal segment of the TM domain (position 3).

(A) Substitution of the KIR TM lysine by arginine (K9R) substantially reduced assembly with DAP12 (lanes 1 and 2). No assembly was observed with the Fcγ signaling dimer (lanes 3 and 4).

(B) Substitution of the FcαRI receptor TM arginine by lysine (R3K) substantially reduced assembly with Fcγ (lanes 1 and 2). No assembly was observed with the DAP12 signaling dimer (lanes 3 and 4). SDS-PAGE was performed under nonreducing conditions. DAP12-associated KIR (A) or Fcγ-associated FcαRI (B) quantitations were normalized to total translation levels of these proteins.
