Rac3-induced neuritogenesis requires the GTPase activity and the membrane binding of Rac3, whereas it is inhibited by the overexpression of HANeurabin I lacking the F-actin or the PDZ domains. (A) FITC-conjugated anti-FLAG immunofluorescence on SK-N-BE cells transfected with FlagRac3V12 (V12), FlagRac3N17 (N17), FlagRac3ΔC-ter (ΔC-ter), or FlagRac3 (WT) and analyzed by confocal microscopy. Note that only wild-type FlagRac3 stimulates the extension of long and branched neurites. (B) FlagRac3 vector (FlagRac3) was cotransfected in SK-N-BE cells together with the indicated HANeurabin I vectors coding for wild type (HANeurabin I) or F-actin (HAΔF-actin), PDZ (HAΔPDZ), or coiled-coil domain (HAΔ511-721) deletion mutants. The same fields of cells immunostained with FITC-conjugated anti-FLAG (top) and with TRITC-conjugated anti-HA antibodies (bottom) are shown. Note that only wild-type HANeurabin I and HANeurabin I Δ511-721 allow the FlagRac3-induced neuritogenesis. Bar, 10 μm.
