Hsf4b interacts with both ERK1 and DUSP26. H1299 cells were transiently transfected with plasmids containing GST alone or GST-Hsf4b (196 to 493) and cotransfected with plasmids encoding HA-ERK1 and Flag-DUSP26 as indicated. At 48 h after transfection, cell lysates were subjected to immunoprecipitation experiments using antibody to Flag-DUSP26 (left panels). The immunoprecipitated proteins were subjected to immunoblotting using antibodies to HA to detect ERK1 or to GST to detect GST alone or GST-Hsf4b. Right panels presents immunoblotting of cell lysates to detect the expression of HA-ERK1, Flag-DUSP26, and GST or GST-Hsf4b using antibodies to HA, Flag, or GST, respectively.
