FLRT3-mediated cell sorting is calcium dependent. 293T cells were co-transfected with enhanced green fluorescent protein (EGFP) and either empty vector (control) or V5-tagged Xenopus FLRT3. 24 h after transfection, cells were dissociated, mixed 1:2 with non-transfected cells and re-plated on plastic or agarose in Ca²⁺-free DMEM supplemented with 10% FCS (a source of ∼ 0.2 mM Ca²⁺ final concentration) and the indicated final concentration of CaCl₂. Pictures were taken after 24 h (plastic, confluent cells) or 68 h (agarose). Scale bars, 200 μm.
