Selective blockage of LAMP-2A expression in mouse fibroblasts. Expression of LAMP-2A was reduced in mouse fibroblasts in culture by using a vector-based method for stable RNAi. Three different regions of the LAMP-2 exon 8A were targeted by using three different vectors, and clones stably transfected with each of the vectors [LAMP-2A(−) cells] were isolated by antibiotic selection. (A) Immunoblot for LAMP-2A, -B, and -C and LAMP-1 of total cellular extracts (150 μg of protein) of WT cells or three LAMP-2A(−) clones each carrying RNAi against a different region of LAMP-2A (c1, c2, and c3). (B) Comparison of total levels of LAMP-2A, -B, and -C mRNA in the same cells. Values are expressed as mRNA amount relative to the values in the control cells (arbitrary value of 1) and are the mean + SE of three different clones containing the construct against the intermediate region of exon 8a (c2).
