Fig. 3. — K56ac is absent in asf1Δ mutants. (A) Western blot analysis of histone H3 K56ac in chromatin assembly factor mutant strains. Whole-cell extracts of the indicated strains were analyzed by using the K56ac antibody; an anti-H3 C terminus antibody was used to detect total histone H3 levels. Recombinant H3 (rH3) and the K56R mutant strain were analyzed simultaneously to demonstrate the specificity of the K56ac antibody. OB, strains purchased from Open Biosystems in the BY4741 background; HS, strains derived from the histone shuffle strain MSY421. (B) The indicated anti-acetyl histone antibodies were used to analyze the indicated OB-strain WT and asf1Δ extracts.
