The EGFR-HER3 interaction. The extent of EGFR phosphorylation in response to EGF and NRG are plotted for cases when EGFR is coexpressed with (A) HER3 alone in the absence of HER2 and (B) varying amounts of HER2 in the presence of a constant amount of HER3. Varying the levels of HER2 and HER3 has a minimal effect on the extent of EGFR phosphorylation. Whereas increasing HER2 causes a slight increase in EGFR phosphorylation, increasing HER3 slightly decreases EGFR phosphorylation in response to EGF. The NRG response is evidence of cross-activation of EGFR either by ligand-bound HER3 (A) or by HER2 activated by ligand-bound HER3 (panel B).
