TABLE 1.
Effect of tyrosine kinase inhibitors on O2− generation induced by alpha-toxina
| Treatment | Inhibitor concn (nM) | Superoxide generation (% of control response) |
|---|---|---|
| Control (alpha-toxin alone) | 100 | |
| K252a | 1.0 | 81 ± 3 |
| 5.0 | 52 ± 6* | |
| 10 | 23 ± 4* | |
| AG1478 | 1.0 | 98 ± 3 |
| 5.0 | 103 ± 2 | |
| 10 | 97 ± 4 | |
| PP2 | 1.0 | 102 ± 3 |
| 5.0 | 100 ± 2 | |
| 10 | 99 ± 4 |
Rabbit neutrophils (1.0 × 106 cells/ml) were preincubated with various concentrations of K252a, PP2, or AG1478 at 37°C for 60 min, and the cells were incubated with 1.0 μg/ml alpha-toxin at 37°C for 3 min. Data are expressed as a percentage of the control (generation of O2− induced by alpha-toxin). Superoxide generation values are means ± standard error for five or six experiments. *, P < 0.01, compared with generation of O2− induced by alpha-toxin alone.
