Cells were loaded with DCF and treated with TGF-β (10 ng/ml, 5 min) and imaged using live confocal microscopy. Representative photographs were taken from 3 separate experiments. B: to quantitate ROS production, the Amplex Red assay was used to measure H2O2 production in HUVEC following TGF-β treatment alone and in the presence of DPI, LacZ adenovirus, and dominant-negative mutant Nox4. There were 6 replicates per condition and 3 separate experiments were performed.
