Spatial localization of PBEF expression in canine lung. Panel A represents background staining with only secondary antibodies of injured/ventilated canine lungs. Panel B demonstrates mild sporadic staining of PBEF in uninjured/ventilated canine lung detected by a rabbit anti-canine PBEF affinity purified polyclonal antibody followed by a goat anti-rabbit IgG conjugated with Alexa Fluor 488 (green color). Panel C demonstrates significant staining of PBEF (green color) in injured/ventilated canine lung with positively stained endothelial cells identified by a rabbit anti-human von Willebrand Factor followed by a goat anti-rabbit IgG conjugated with Texas Red (C, red color). Blue color indicates nuclei staining by DAPI. Pre-immune serum did not show any significant staining of PBEF in an injured/ventilated canine lung (data not shown). Photos were at 400 × magnifications.
