Detergent treatments of melon microsomal membranes demonstrate that CmERS1 is an integral membrane protein. Microsomes were prepared from melon leaves. The membrane pellet was resuspended in 0.1 m NaCl, 0.1 m Na2CO3 (pH 11), 1% (v/v) Triton X-100, or 1% (v/v) Sarkosyl. After incubation at 4°C for 1 h, the samples were separated into supernatant (S) and pellet (P) fractions by centrifugation at 125,000g for 1 h. Equal amounts of the proteins (15 μg) were subjected to western-blot analyses with anti-CmERS1 antibody. Numbers on the right indicate the molecular mass.
