Sucrose density gradient centrifugation analysis. Cell lysates were prepared from yeast strain Y1948 containing 3Xmyc::CHS5 and SBE2::3XHA alleles and fractionated in 10–65% sucrose density gradients by centrifugation for 20 h. Fractions (abscissa in A) were collected from the bottom of the gradient and analyzed by immunoblot analysis with the use of antibodies to HA, c-myc, Pma1p, Anp1p, and CPY. Only the vacuolar isoform of the CPY protein (CPYvp) is represented. Sample blots are shown in B. The amount of protein in each lane was quantified, and the results are shown in A as percentages of the total protein.
