Identification of the N-terminal–addressing domain of Aip3p. Wild-type strain FY23×86 was transformed with plasmid pDA259 (A), plasmid pHJ9 (B), plasmid pHJ6 (C), and pHJ34 (D). GFP-aip3p localization was visualized by fluorescence microscopy (A—D, left side), and the same cells were visualized by DIC microscopy (A—D, right side). The domain structure of Aip3p is displayed in E and compared with regions tested for localization as GFP fusions and regions previously reported as displaying two-hybrid interactions (Amberg et al., 1997; Evangelista et al., 1997; Roemer et al., 1998).
