(A) Survival rate of double stable transfectants of Ba/F3 cells established by using retrovirus vectors carrying IRS-1-IRES-Bcl-2, IRS-3-HA-IRES-Bcl-2,or IRS-1-IRES-IRS-3, compared with those of Bcl-2, IRS-1, and Ba/F3 cells. (B) Stable transfectants expressing IRS-1, IRS-2, IRS-3, and mock cells were serum starved and stimulated with insulin (100 nM). Lysates from these cells were subjected to immunoblotting with 4G10. β chain, β subunit of the insulin receptor. (C) Coexpression of the loop fragment of Bcl-2 in Bcl-2 and IRS-1 cells severely impairs the survival of these cells.
