gdt1 cells are not impaired in folate sensing. Ax2 and L8 cells were grown in axenic medium in the presence of 1 mM folate; cell density was kept at or below 106 cells/ml. Samples were removed and prepared for Western blots at the times indicated. In wild-type cells, folate reduced discoidin to undetectable levels within 19 h. Even though L8 cells still showed a strong staining after 24 h of growth with folate, the amount of discoidin was significantly reduced, thus indicating that the mutant could sense and respond to the inhibitory signal. Note that staining in L8 is saturated in the first time points.
