Nascent synthesis of Rubisco LSU in R. rubrum is inhibited by exposure to atmospheric air. Pulse labeling of the photosynthetic bacterium R. rubrum was performed on midlog phase cultures growing at 30°C under anaerobic conditions and after exposure to atmospheric air. Pulses of 5 min were given at intervals of 0, 2, and 4 h with 75 μCi of [35S]Met cell-labeling mix. Equal protein loads that also contained equal amounts of incorporated radioactivity were analyzed over 7.5% to 12.5% SDS-PAGE. The gels were blotted and the blots were reacted with antibodies against global Rubisco. The colored developed blots were aligned with the autoradiogram that is shown in the figure.
