To construct the E5XCM− HPV16 mutant, a single nucleotide, adenine, was deleted in codon 10 of the E5 ORF, disrupting the XcmI restriction site (underlined) and leading to a frameshift with a stop codon (TAA) encountered at codon 28. Only the first 10 amino acids of E5XCM− are derived from the E5 ORF.
