Exogenous XChk1 lengthens cleavage cycles. (A) Embryos were microinjected at the one-cell stage with buffer or 12 ng of FLAG-tagged XChk1 mRNA and photographed when controls reached stage 8 (the MBT). (B and C) Embryos were microinjected at the one-cell stage with 2 ng of FLAG-tagged luciferase (control) mRNA, FLAG-tagged catalytically inactive XChk1 mRNA (NA XChk1), or FLAG-tagged wild-type XChk1 mRNA (XChk1). Embryos were photographed when controls reached stage 7 (B; pre-MBT) and stage 9 (C; post-MBT). (D) Western analysis of XChk1 protein in embryos treated and collected as in B and C. NA XChk1, inactive XChk1 mRNA; wt XChk1, wild-type XChk1 mRNA. Numeric values represent relative band intensity as determined by densitometry.
