5′ RACE used to determine the transcriptional initiation site of the SAK gene. (A) Sequence demonstration of transcription initiation site of the SAK gene. 5′ RACE was performed with Ambion's FirstChoice RLM-RACE kit according to the manufacturer's instructions. The transcription initiation site was determined by comparing the sequences immediately after the 5′ RACE adapter (5′-GAACACTGC GTTTGCTGGCTTTGATGAAA-3′) to the SAK gene sequence through the evidence viewer from NCBI. The SAK mRNA start was indicated. (B) DNA sequence of -171 to +250 of the SAK gene.
