Metabolically labeled E3-13.7 proteins and EGF receptors follow divergent pathways subsequent to early endosomes. ERwt cells that had been infected with in724 were pulse-labeled for 15 min at 15 h after infection, incubated in chase medium for 45 min or 180 min, and then processed for Percoll gradient cell fractionation. Five fractions (I–V) were collected starting from the top of the gradient. Membranes were solubilized with RIPA detergent and sequentially immunoprecipitated with antibodies to E3-13.7 and the EGF receptor. Immunoprecipitates were resolved by SDS-PAGE and detected by fluorography.
