Molecular Cloning of the REP1 Gene.
(A) Genomic structure of the REP1 gene. The locations of start codon (ATG) and stop codon (TGA) are shown. The triangles indicate the positions of T-DNA insertions in the rep1-1 and rep1-2 mutants, respectively. The structure of cDNA is shown along with the regions for the bHLH motif. (C) RNA gel blot analysis of rep1 mutants. Total RNA was extracted from seedlings of the wild type and rep1-1 and rep1-2 mutants that had been grown in darkness for 4 days and then subjected to RNA gel blot analysis with a REP1 cDNA probe. Ten micrograms of total RNA was loaded onto each lane. The 18S rRNA probe was used as a loading control.
