Promoter constructs carrying mutations through various response elements (see Fig. 1) were cotransfected with either CMV-GH (black bars) or pCMVhAR (hatched bars). Transfections were as described in the legend of Fig. 2 where relative activity represents the luciferase/β-galactosidase activity of each promoter made relative to the wild type promoter co-transfected with CMV-GH. The relative activity of each construct in the presence and absence of AR was used to calculate the percent repression (inset).
