Effects of mutant Sso1p TMD when appended to Snc1p. GFP-tagged constructs consisting of Snc1p with the Sso1p TMD (SSSO), with Asp residues at position 3, 5, 9, or 13 as indicated, were expressed in wild-type (wt) or Δrer1 mutant cells, and confocal images were obtained. Much of the fluorescence is in vacuoles, but arrowheads indicate nuclear envelope staining with the 3D and 5D mutants (nuclei being identified by DAPI staining of the cells). Fainter fluorescence corresponding to peripheral ER was also visible in these cells.
