(A) In vivo incorporation of [³⁵S]methionine in wild-type and Δint6 fission yeast cells. Total incorporation of [³⁵S]methionine into proteins in a wild-type or Δint6 strain was determined. (B) Analysis of polysomes. Cell lysates were prepared from exponentially growing cultures of a wild-type strain (left) or a Δint6 strain (right) in YEA medium at 32°C and subjected to 7–47% (wt/vol) sucrose gradient as described in MATERIALS AND METHODS. Each gradient was fractionated in an ISCO gradient fractionator, and the absorbance profile at 254 nm was analyzed in an ISCO UA-5 absorbance monitor.
