Pattern of development of anti-PLC-β1 immunostaining in isolated nuclei. The selected photographs are representative of the development of the pattern of anti-PLC-β1 immunostaining observed by confocal microscopy in isolated nuclei. The nuclei were isolated from oocytes cultured for 0, 30, 60, or 90 min and immediately immunostained with the anti-PLC-β1 mAb. Intense labeling around the nuclear membrane; the nucleoplasm was very slightly stained at time zero. Very bright nuclear membrane and slight labeling in the nucleoplasm and around the nucleolus after 30 min in culture. Large spots in the nucleoplasm and a less stained nuclear membrane after 60 min in culture. Major immunoreactivity with more fluorescent spots after 90 min in culture. Bar, 10 μm.
