The frequency of IFN-γ–producing donor-specific T cells in C57BL/6 mice following allogeneic cardiac transplantation. Mice were transplanted with either MHC-mismatched BALB/c (left columns, n = 3) or minor-mismatched 129X1/sv (right columns, n = 3) allogeneic hearts. Recipient spleens were removed after rejection, and splenocytes (1.2 × 106 cells per well) were incubated with either recipient (white bars) or donor (black bars) irradiated splenocytes. The frequencies of IFN-γ–producing T cells were then determined using ELISPOT assay as described in Methods. *P = 0.001.
