p104 is essential for launching. (A) Mutations were introduced into the well conserved GDD motif (EFD) or its neighboring amino acid (I497V) of p104. Transformants with launching plasmids having the modified p104 were grown and induced for 23S RNA. RNA was extracted from the induced cells, separated in an agarose gel, and analyzed by hybridization using a 23S RNA positive or negative strand-specific probe.
