Postnuclear supernatants of transiently transfected CHO-K1 cells were subjected to ultracentrifugation in a sucrose gradient after which fractions were collected and Western blotted. Fractions from the top, 1–5; pellet, 6. A minor fraction of ORP1L flotated with total membranes (A), whereas ORP1S was only recovered from the cytosolic fractions (B). The membrane association was most pronounced with the N-terminal fragment of ORP1L containing the ankyrin repeat region and the PHD (C).
