Resistance of hepatocytes from ASMase knockout mice to TNF-α–induced apoptosis. ASMase+/+ (a) or ASMase–/– (b) mouse hepatocytes were first pretreated with HP to deplete the mGSH pool and then treated with either TNF-α (280 ng/ml) or ASMase (0.4 U/ml) for 12 hours. Cell death was determined by Hoechst 33258 and propidium iodide staining. Results are expressed as means ± SD (n = 5 independent experiments). *P < 0.05 versus control.
