Effect of mutations in hImp3, hImp4 and hMpp10 on their subcellular localization. HEp-2 cells were transiently transfected with constructs encoding GFP fusion proteins of hImp3, hImp4, hMpp10 and mutants thereof and the localization of the fusion proteins was determined by fluorescence microscopy. The mutants used in these experiments are outlined in Figures 6A and 7A. The corresponding phase contrast image is shown on the left of each fluorescence image. (A) Results for GFP alone; (BD) GFP-tagged hImp3 and hImp3 mutants; (EI) GFP-tagged hImp4 and hImp4 mutants; (JN) GFP-tagged hMpp10 and hMpp10 mutants. The arrow indicates the nucleolar accumulation of GFPhMpp10Δ565 in a cell expressing relatively low levels of this mutant.
