Calpain inhibition affects neutrophil morphology and adhesion. Neutrophils were incubated for 20 min in CCM-1 containing vehicle (a), 50 μg/ml (130 μM) ALLN (b), 50 μg/ml (125 μM) ALLM (c), 75 μM compound 73 (d), 1 μM PD150606 (e), 79 μM calpastatin peptide (f), 75 μM compound 42 (g), 200 μM cathepsin inhibitor I (h), or 50 μM clasto-lactacystin β-lactone (i). Cells were then plated onto 2.5 μg/ml Fbg for 30 min at 37°C. Morphology was compared with cells treated with 1.25 nM IL-8 (j) or 100 nM fMLP (k) as described in Materials and Methods. All samples were subsequently imaged by time-lapse videomicroscopy. A representative frame is shown. Bar = 50 μm.
