Calpain may act downstream of GPCRs. Neutrophils were pretreated for 1 h in CCM-1 containing 0.5 mg/ml PTx or vehicle at 37°C. Subsequently, 50 μg/ml ALLN or vehicle was added, and cells were plated on 2.5 μg/ml Fbg for 25 min. IL-8 was added during the last 5 min to appropriate dishes, and cells were imaged as in Fig. 4.
