bur6-1 and mot1-301 mutations increased total mRNA levels. Slot blot hybridization to detect total poly(A)⁺ RNA of mot1-301 (lane 2) and its isogenic wild-type strain (lane 1, FY86), and the bur6-ts mutant (lane 4) and its isogenic wild-type strain (YM208, lane 3). Exponentially growing cells in YPD (samples 23°C) were shifted to 37°C and incubated for 90 min. Total RNA was analyzed with a ³²P-labeled poly(dT) probe. rRNA was used to verify the application of equal amounts of sample (our unpublished data).
