Analysis of telomere length in polα and primase mutants. polα and spp1 and spp2 (genes coding for S. pombe primase subunits 1 and 2, respectively) mutants were subcultured for ∼100 generations at their respective semipermissive temperature (polα and spp1 mutants, 25°C; spp2 mutants, 33°C) before harvest. Genomic DNA was restricted with ApaI and separated on 1.2% agarose gels, transferred to Hybond N+ filters, and probed with a 1.9-kb ApaI fragment of pEN42 (55). The positions of DNA size markers and their sizes in base pairs are given on the left. Brackets indicate the weak smear seen in the polαts13 mutant.
