Membrane binding activity of HIS-PLO proteins. HIS-PLO proteins were diluted to a concentration of 2.5 μg/ml in bovine serum albumin (1 mg/ml), 0.5 ml was added to an equal volume of 10% ovine blood, and the mixture was incubated on ice for 20 min. The cells were subsequently harvested by centrifugation and lysed in sodium dodecyl sulfate-polyacrylamide gel electrophoresis sample buffer. Samples incubated with HIS-PLO (lane 1) or HIS-PLO.A₅₂₂ (lane 2) were separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, transferred to nitrocellulose, and immunostained with a 1/100 dilution of goat anti-HIS-PLO. The position of the 55-kDa PLO band is indicated by the arrow. Molecular mass markers, in kilodaltons, are indicated on the left.
