Brain COX-2 mRNA, protein and enzymatic activity levels in wild type and COX-1−/− mice. (A) Brain COX-2 mRNA expression was measured using real-time PCR. COX-2 mRNA levels were normalized to an endogenous reference (pgk1) in the COX-1−/− mice relative to the wild type, using the ΔΔCT method. The results are expressed as mean ± SEM of six independent samples, each assayed in triplicate. (B) Representative immunoblots illustrating COX-2 (72 kDa) and β-actin (42 kDa) protein levels. Relative integrated density values of COX-2 protein level normalized to β-actin protein level in brain from COX-1−/− mice is expressed as percentage of protein levels in wild type mice. Each sample derived from an individual mouse brain (n = 12). *p ≤0.05 versus wild type. (C) Brain COX-2 enzymatic activity in wild type and COX-1−/− mice was assessed colorimetrically using a commercial COX activity assay kit that measures the peroxidase activity of COX.
