Expression of Hc2 during infection. The figure shows the results of Western blot analysis of C. trachomatis L2 Hc2 and major outer membrane protein (MOMP) over a 24-h infection time period. HeLa cells in 24-well plates were infected at a multiplicity of infection of 500 for 30 min at 4°C to allow attachment, the inoculum was removed, and the cultures were fed with prewarmed medium and incubated at 37°C. The time of temperature upshift was considered time zero. The cultures were solubilized at the times indicated, and equal amounts were loaded for sodium dodecyl sulfate-polyacrylamide gel electrophoresis and immunoblotting. “UI” indicates uninfected sample, and “C” (control) represents samples collected from a 24-h infection in the continuous presence of chloramphenicol. Results shown are representative of three separate experiments.
